Abstract: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental condition characterized by impairments in social communication, repetitive behaviors, and heightened sensitivity to stress. In recent years, increasing evidence has highlighted the significance of the gut–brain axis, which reflects the functional interaction between the gut microbiota and the central nervous system, in the pathophysiology of ASD. This article examines the psychobiotic potential of Lactobacillus strains in modulating ASD-associated behavioral disturbances. The study discusses the modulation of gut–brain axis signaling, the effects on hypothalamic–pituitary–adrenal (HPA) axis activity, reductions in cortisol levels, and attenuation of stress reactivity. In addition, the influence of Lactobacillus strains on the expression of brain-derived neurotrophic factor (BDNF), as well as their capacity to synthesize ?-aminobutyric acid (GABA), short-chain fatty acids (SCFAs), and metabolites involved in dopamine signaling, is analyzed. Furthermore, the effects of Lactobacillus strains on the serotonergic system via tryptophan metabolism and their immunomodulatory properties are highlighted. The findings indicate that Lactobacillus strains represent a promising psychobiotic approach as an adjunctive strategy for ASD management.
